AC OleaShield

Fibroblasts incubated with AntA, a known inducer of oxidative stress, elicited a 57% increase in ROS levels, compared to untreated fibroblasts. These data demonstrate the supraphysiologic level of ROS induced by AntA and the magnitude of ROS in fibroblasts is dynamic. Conversely, fibroblasts treated with AC OleaShield at 0.01%, 0.1%, and 1.0% demonstrated 18%, 28%, and 35% reductions in ROS levels compared to fibroblasts treated with AntA, respectively. These data demonstrate AC OleaShield attenuates excessive oxidative stress. Collectively, intrinsic and extrinsic factors perturb skin homeostasis by stimulating abundant levels of ROS that amplify DNA mutation, cellular senescence, advanced glycation end products, protein oxidation, and collagen degradation. These data indicate AC OleaShield scavenges unnecessary ROS, which may help to attenuate characteristics of cellular aging.

AC OleaShield was able to increase cell migration and close the scratch at a rate comparable to the positive control. The mechanisms of the cells in the in vitro scratch assay mimic the mechanisms seen in in vivo wound healing therefore we can be assured that our results are translatable outside the laboratory. With the present study we can be confident that this product has healing abilities and cell proliferation properties.

Treated with AC OleaShield at 0.01%, 0.1%, and 1.0% demonstrated 13%, 19%, and 38% increases in autophagic-lysosomal vacuoles compared to untreated fibroblasts, respectively. These data demonstrate AC OleaShield activates fibroblast detoxification. Collectively, autophagy is a biological process that identifies toxins within the cell and removes or recycles damaged cellular components to maintain skin homeostasis. These data indicate AC OleaShield triggers this cellular detoxification process, which may help to attenuate characteristics of cellular aging.

AC OleaShield (12006) was tested to evaluate its effects on the viability of normal human dermal fibroblasts (NDHF). At concentrations of 0.1% and 0.01%, AC OleaShield, nor the preservatives contained therein exhibited any inhibition of cell viability. It can therefore be concluded that at normal use concentrations AC OleaShield is not cytotoxic.

AC OleaShield exhibited greater antioxidant activity than 200μM Trolox®. The antioxidant capacity of AC OleaShield increased as the concentration increased. As a result, we can assure that its ability to minimize oxidative stress is dose dependent. Maximizing the antioxidant capacity on a cellular level allows for ROS to be dealt with at a rate that provides protection from cellular damage. This cellular damage can be seen as physical signs of aging such as wrinkles, loss of elasticity, unwanted pigmentation, and skin unevenness with slow regeneration.

AC OleaShield exhibited greater antioxidant activity than 12.5 μM Trolox. The radical scavenging activity of AC OleaShield increased as the concentration increased, indicating the antioxidant properties of AC OleaShield are dose dependent. Provided the nitrogen radical of DPPH is well-protected, these results demonstrate the strong ability of AC OleaShield to scavenge harmful radicals that are not easily reduced. Collectively, quenching excessive radicals with exogenous assistance preserves cellular homeostasis and blunts intrinsic and extrinsic age-related declines in antioxidant capacity. These data indicate AC OleaShield is capable of scavenging well-protected radicals, which may help to attenuate characteristics of cellular aging.

As evidenced in this four-week study, AC OleaShield is capable of reducing the appearance of fine lines on the face. After four weeks, participants applying 5.0% AC OleaShield demonstrated a 6% decrease in the overall number of wrinkles and a 2% reduction in the size and intensity (score) of those wrinkles, compared to baseline. After treatment ended, the size, intensity, and number of wrinkles in participants applying 5.0% AC OleaShield continued to outperform the base lotion alone. After two weeks of regression, the participants that applied 5.0% AC OleaShield demonstrated a 7% reduction in the total number of wrinkles, while the base lotion produced an increase of 2%.

As evidenced in this four-week study, AC OleaShield is capable of significantly reducing the appearance of porphyrins on the face. After four weeks, participants applying 5.0% AC OleaShield demonstrated an 11% decrease in the overall number of porphyrins, compared to baseline. Conversely, base lotion application increased the total number of porphyrins by 12% in four weeks. These results indicate that applying 5.0% AC OleaShield for four weeks provides a reduction of porphyrin appearance on the face, resulting in a more youthful skin appearance. After treatment ended, the reduction of porphyrins for participants applying 5.0% AC OleaShield continued to outperform the base lotion alone. After two weeks of regression, the participants that applied the 5.0% AC OleaShield demonstrated a 22% reduction in the total number of porphyrins.
